Influence of Diazepam on Blood Glucose Levels in Nondiabetic and Non-Insulin-dependent Diabetic Subjects Under Dental Treatment With Local Anesthesia. METHODS

Ethical approval for this investigation was obtained from the Ethical Committee in Research at the Piracicaba Dental School/University of Campinas (UNICAMP). Twenty volunteers took part in this study: 10 *non-insulin-dependent subjects (mean age 48.4 ± 6.65 years) and 10 healthy subjects (mean age 41.9 ± 11.67 years).

The diabetic volunteers should have presented glycosylated hemoglobin <10% and fasting blood glucose <180 mg/dL. The volunteers were instructed to attend the sessions after an overnight fast (8 hours) and not alter the schedule of their usual medications. The study protocol was as follows. Each volunteer attended on 3 occasions, at the same time of the day, exactly 3 weeks apart. The volunteers were allowed to take a meal in the previous day until 11 pm.

First Session

At 7 am a cannula was inserted into an antecubital vein, and this was kept patent with a flush of heparinized saline. Two fasting blood samples were taken for estimation of blood glucose level and glycosylated hemoglobin. At 60, 80, 95, 125, and 155 minutes after the breakfast, blood samples were taken for estimation of blood glucose level. These were the baseline samples. Second and Third Sessions

As in the first session at 7 am, a cannula was introduced into an antecubital vein and maintained with a flush of heparinized saline. A fasting blood sample was taken for estimation of blood glucose level followed by breakfast and the intake of 5 mg diazepam or placebo. These capsules were administered in a random order and double-blind manner. Sixty minutes after breakfast, blood samples were taken and the volunteers were submitted to an infiltration of 1.8 mL of 2% mepivacaine with 1: 100,000 adrenaline (Mepivacaine) injected over a period of 2 minutes in the incisive region of the lower jaw.
